Cell and Molecular Biology Laboratory

Course Objective:

The course aims to,

  • understand the principle of nucleic acid isolation.
  • understd the principles of PCR and their uses in genetic engineering.
  • gain a thorough knowledge about nucleic acid hybridization.
  • learn the history of DNA sequencing and current methods and gene synthesis

  1. Preparation of Genomic DNA
  2. PCR amplification of gene from the genomic DNA
  3. Preparation of plasmid DNA
  4. Detection of Plasmid DNA by Agarose gel electrophoresis
  5. Restriction Digestion of the vector and Insert
  6. Ligation and Transformation ofE.coli
  7. Lysate PCR confirmation.
  8. Restriction and gel elution of DNA fragments
  9. Electroporation of Yeast
  10. SDS-PAGE analysis of purified protein
  11. Western blot confirmation of expressed protein (anti his)
  12. Quantification of proteins.
  13. RNA Isolation
  14. cDNA preparation from RNA
  15. Site directed mutagenesis
  16. Southern hybridization experiment

REQUIRED FOR 30 STUDENTS

  • PCR machine
  • Electrophoretic assemblies for DNA and protein separations
  • ELISA reader
  • Ultracentrifuge
  • Laminar air flow cabinets
  • Cooling centrifuge

Course Outcome:

At the end of the course the students will be able to

  1. experience basic techniques of DNA isolation and manipulation
  2. experience in selecting genetically transformed organisms for downstream analysis
  3. experience basic techniques involved in analysis of gene expression at nucleic acids and proteins level
